Analytical Data

  • Gene name

    RSPO3/R-spondin-3

  • 简介

RSPO3 is a potent activator of the canonical Wnt pathway and can bind to LGR4-6 receptors to initiate a complex with phosphorylated LRP6 and Frizzled receptors. This interaction activates the canonical Wnt pathway and upregulates target genes. R-spondin 3 Protein, Human (HEK293, His) is the recombinant human-derived R-spondin 3 protein, expressed by HEK293 , with C-His labeled tag.

  • Application

    SPRMSTBLIITCELISACELL ASSAYDRUG SCREENING

  • Biological Activity

    1. Measured by the ability to enhance BMP-2 induced alkaline phosphatase activity in MC3T3-E1 cells. The ED50 for this effect is 1.787 μg/mL . 2. Measured by its ability to induce Topflash reporter activity in HEK293T human embryonic kidney cells. The EC50 for this effect is 20ng/ml in the presence of 5ng/mL Recombinant Human Wnt-3a. Measured by the ability to enhance BMP-2 induced alkaline phosphatase activity in MC3T3-E1 cells. The ED50 for this effect is 1.787 μg/mL. Corresponding to a specific activity is 559.597 U/mg.

  • Stability Test

The thermal stability is described by the loss rate. The loss rate was determined by accelerated thermal degradation test, that is, incubate the protein at 37℃ for 48h, and no obvious degradation and precipitation were observed. The loss rate isless than 8% within the expiration date under appropriate storage condition.

  • Storage & Shelf Life

Samples are stable for up to twelve months from date of receipt at -20℃ to -80℃. Store it under sterile conditions at -20℃ to -80℃. It is recommended that the protein be aliquoted for optimal storage. Avoid repeated freeze-thaw cycles.

  • Shipping

In general, recombinant proteins are supplied as lyophilized powder and shipped at ambient temperature. For bulk packages, the proteins are provided as frozen liquid and shipped with blue ice, unless otherwise requested by the customer.

Protein Description

R-spondin-3 (RSPO3) is a member of the R-spondin family of secreted proteins that play critical roles in regulating Wnt signaling pathways, which are important for a variety of developmental processes and tissue homeostasis. The significance of RSPO3 has been highlighted in studies related to embryonic development, organogenesis, and cell proliferation, particularly in the context of stem cell biology and cancer. Research has shown that RSPO3 can enhance Wnt signaling by binding to LGR4-6 receptors, thus promoting the proliferation of stem cells in various tissues, including the intestine and skin. Furthermore, irregularities in RSPO3 expression have been implicated in several diseases, such as colorectal cancer and other malignancies. As a result, RSPO3 and its signaling mechanisms have emerged as potential therapeutic targets. The recombinant production of RSPO3 facilitates detailed investigations into its biological functions and interactions with Wnt signaling components. Understanding the molecular mechanisms governed by RSPO3 can potentially lead to the development of novel strategies for cancer treatment and regenerative medicine, particularly by harnessing its ability to modulate stem cell activity and tissue regeneration.
